The tissue is rinsed in the residue of ‘hematoxylin’ vigorously in warm water. After this step, two to three drops of eosin stains are put on the tissue to seek the intended difference within thirty seconds. Again, the tissue is rinsed in the stain with cold water and finally, dipped in ethanol fixate, then in xylene for forty-five seconds (Yakovlev, 2009).

Results: Blue nuclei, with some metachromasia and the cytoplasm, contain several shades of pink that detect the presence of different tissue components (Luna, 1960).
Sources of Error: Eosin Leeching and/or Inadequate Staining

Principle of the Technique: Cutting paraffin section by four micron

Procedure (Steps): Bouin’s solution, an inorganic oxide, is used in the microwave for one minute and is allowed to stand for fifteen minutes. Subsequently, for five minutes, the stain is washed in tap water, followed by the application of one drop of hematoxylin, and is allowed to stand for ten minutes. Correspondingly, it is again washed in tap water for five minutes and then rinse in distilled water. In the following step, the stain is dipped in Dietrich scarlet solution for five minutes and then rinsed in distilled water.
